Definition | The Chromium Controller is a single cell gene sequencer and immunity profiler. It uses advanced microfluidics to perform single cell partitioning and barcoding on a timescale of minutes. The instrument utilises gel beads, each containing millions of barcoded oligonucleotides, which are mixed with cells or nuclei from a sample pool at a flow rate designed to enable each gel bead to bind to a single cell or nucleus. Once bound, the gel beads are immersed in an oil solution, capturing each single bead, cell, and the immediately surrounding reagents in a bubble which then serves as a reaction vesicle. Using advanced microfluidics, the instrument encapsulates the gel beads in GEMs (gel beads-in-emulsion), along with a single cell or nucleus and reagents to create a micro reaction. The Chromium Controller enables large scale integrated multiomic analysis, with a cell throughput of 500 to 140,000 singlets per run. The instrument allows for several assays - single cell gene expression, immune profiling, Assay for Transposase-Accessible Chromatin (ATAC), and multiome ATAC + gene expression. | ||
